PTH analogs oral
Human Parathyroid Hormone(PTH) is an 84 amino acid peptide hormone which is involved in the regulation of calcium and phosphate metabolism in the bones and kidney. Paradoxically, PTH can cause opposing skeletal effects depending on the pattern of exposure to the hormone. Chronic elevation of PTH results in a greater degree of osteoclastic bone resorption. In contrast, intermittent administration of PTH has the effect of enhancing bone formation, by stimulating osteoblast activity to a greater degree than osteoclast activity. PTH is the first hormone that has been confirmed to have a potent anabolic effect in patients with established osteoporosis and the first anabolic product approved in the U.S. and Europe for the treatment of severe osteoporosis. This increase in bone growth results in a significant decrease in vertebral and total body fracture rates. The anabolic activity can be demonstrated by intermittent administration of the entire hormone, or N-terminal fragments and analogs of the hormone.
